    Efficient bypass mechanisms for low latency networks on-chip

    RESUMEN: La importancia de las redes en-chip en los procesadores multi-núcleo es cada vez mayor. Los routers con baipás son una solución eficiente para reducir la latencia de estas redes. Existen dos tipos de redes con baipás: single-hop y multi-hop. Las redes con baipás single-hop minimizan la latencia individual de cada router al asignar los recursos del router con antelación a la recepción de los paquetes. Las redes con baipás multi-hop, conocidas como SMART, permiten que los paquetes atraviesen múltiples routers en un único ciclo. La primera propuesta de esta tesis es Non-Empty Buffer Bypass (NEBB), un mecanismo que incrementa la utilización del baipás de tipo single-hop, eliminando la necesidad de usar canales virtuales. Para redes con baipás multi-hop propone SMART++ y S-SMART++. SMART++ elimina la necesidad de SMART de usar una gran cantidad de canales virtuales para aprovechar el ancho de banda de la red, permitiendo el diseño de configuraciones de bajo coste. S-SMART++ hace uso de la asignación de recursos de forma especulativa para preparar el baipás de tipo multi-hop. Este mecanismo reduce la latencia y su dependencia con la longitud máxima de los saltos de tipo multi-hop, aspecto clave para su viabilidad en diseños reales. La contribución final es un conjunto de herramientas de código abierto llamada Bypass Simulation Toolset (BST) compuesto por versiones extendidas de BookSim y OpenSMART, una API para integrar BookSim en otros simuladores y una serie de scripts para facilitar el diseño y evaluación de este tipo de redes.ABSTRACT: Networks on-Chip (NoCs) are becoming more important in many-core processors as the number of cores grows. Bypass routers are an efficient solution that skips pipeline stages. There are two types of bypass mechanisms: single-hop and multi-hop bypass. Single-hop bypass minimizes the router delay by skipping allocation stages in each hop. Multi-hop bypass, called SMART, minimizes the effective number of hops by traversing multiple routers in a single cycle. The first proposal of this dissertation is Non-Empty Buffer Bypass (NEBB) for single-hop bypass, which increases the bypass utilization without requiring VCs to match traditional bypass routers. It proposes SMART++ and S-SMART++ for multi-hop bypass. SMART++ removes the requirement of using multiple VCs of SMART to exploit the bandwidth of the network, enabling low-cost configurations. S-SMART++ relies on speculative allocation to set up multi-hop bypass paths. Thus, it reduces latency and its dependency with the maximum length of multi-hops, relaxing the requirements to integrate multi-hop bypass in real designs.     S-SMART++: A Low-Latency NoC Leveraging Speculative Bypass Requests

    Many-core processors demand scalable, efficient and low latency NoCs. Bypass routers are an affordable solution to attain low latency in relatively simple topologies like the mesh. SMART improves on traditional bypass routers implementing multi-hop bypass which reduces the importance of the distance between pairs of nodes. Nevertheless, the conservative buffer reallocation policy of SMART requires a large number of Virtual Channels (VCs) to offer high performance, penalizing its implementation cost. Besides, SMART zero-load latency values highly depend on HPC Max HPCMax, the maximum number of hops that can be jumped per cycle. In this article, we present Speculative-SMART++ (S-SMART++), with two mechanisms that significantly improve multi-hop bypass. First, zero-load latency is reduced by speculatively setting consecutive multi-hops. Second, the inefficient buffer reallocation policy of SMART is reduced by combining multi-packet buffers, Non-Empty Buffer Bypass and per-packet allocation. These proposals are evaluated using functional simulation, with synthetic and real loads, and synthesis tools. S-SMART++ does not need VCs to obtain the performance of SMART with 8 VCs, reducing notably logic resources and dynamic power.     Lagarto I RISC-V Multi-core: Research Challenges to Build and Integrate a Network-on-Chip

    Current compute-intensive applications largely exceed the resources of single-core processors. To face this problem, multi-core processors along with parallel computing techniques have become a solution to increase the computational performance. Likewise, multi-processors are fundamental to support new technologies and new science applications challenges. A specific objective of the Lagarto project developed at the National Polytechnic Institute of Mexico is to generate an ecosystem of high-performance processors for the industry and HPC in Mexico, supporting new technologies and scientific applications. This work presents the first approach of the Lagarto project to the design of multi-core processors and the research challenges to build an infrastructure that allows the flagship core of the Lagarto project to scale to multi- and many-cores. Using the OpenPiton platform with the Ariane RISC-V core, a functional tile has been built, integrating a Lagarto I core with memory coherence that executes atomic instructions, and a NoC that allows scaling the project to many-core versions.     DNA Vaccination Partially Protects against African Swine Fever Virus Lethal Challenge in the Absence of Antibodies

    The lack of available vaccines against African swine fever virus (ASFV) means that the evaluation of new immunization strategies is required. Here we show that fusion of the extracellular domain of the ASFV Hemagglutinin (sHA) to p54 and p30, two immunodominant structural viral antigens, exponentially improved both the humoral and the cellular responses induced in pigs after DNA immunization. However, immunization with the resulting plasmid (pCMV-sHAPQ) did not confer protection against lethal challenge with the virulent E75 ASFV-strain. Due to the fact that CD8+ T-cell responses are emerging as key components for ASFV protection, we designed a new plasmid construct, pCMV-UbsHAPQ, encoding the three viral determinants above mentioned (sHA, p54 and p30) fused to ubiquitin, aiming to improve Class I antigen presentation and to enhance the CTL responses induced. As expected, immunization with pCMV-UbsHAPQ induced specific T-cell responses in the absence of antibodies and, more important, protected a proportion of immunized-pigs from lethal challenge with ASFV. In contrast with control pigs, survivor animals showed a peak of CD8+ T-cells at day 3 post-infection, coinciding with the absence of viremia at this time point. Finally, an in silico prediction of CTL peptides has allowed the identification of two SLA I-restricted 9-mer peptides within the hemagglutinin of the virus, capable of in vitro stimulating the specific secretion of IFNγ when using PBMCs from survivor pigs. Our results confirm the relevance of T-cell responses in protection against ASF and open new expectations for the future development of more efficient recombinant vaccines against this disease

    Association of APOA5 and APOC3 genetic polymorphisms with severity of hypertriglyceridemia in patients with cutaneous T-Cell lymphoma treated with bexarotene

    Importance: hypertriglyceridemia is the most frequent and limiting adverse effect of bexarotene therapy in cutaneous T-cell lymphoma (CTCL). Despite standard prophylactic measures, there is a wide variability in the severity of this complication, which could be associated with both genetic and environmental factors. Objectives: to analyze the association between genetic polymorphisms of apolipoprotein genes APOA5, APOC3, and APOE and the severity of hypertriglyceridemia during bexarotene therapy and to optimize patient selection for bexarotene therapy based on adverse effect profile. Design, Setting, and Participants: this case series study was conducted in 12 university referral hospitals in Spain from September 17, 2014, to February 6, 2015. One hundred twenty-five patients with a confirmed diagnosis of CTCL who had received bexarotene therapy for at least 3 months were enrolled. Nine patients were excluded owing to missing analytic triglyceride level data, leaving a study group of 116 patients. Data on demographic and cardiovascular risk factor were collected, and a complete blood analysis, including lipid profile and genetic analysis from a saliva sample, was performed. Main Outcomes and Measures: primary outcomes were the maximal triglyceride levels reported in association with the minor alleles of the polymorphisms studied. Results: among 116 patients, the mean (SD) age was 61.2 (14.7) years, 69 (59.5%) were men, and 85 (73.2%) had mycosis fungoides, the most prevalent form of CTCL. During bexarotene therapy, 96 patients (82.7%) experienced hypertriglyceridemia, which was severe or extreme in 8 of these patients (8.3%). Patients who carried minor alleles of the polymorphisms did not show significant differences in baseline triglyceride concentrations. After bexarotene treatment, carriers of at least 1 of the 2 minor alleles of APOA5 c.-1131T>C and APOC3 c.*40C>G showed lower levels of triglycerides than noncarriers (mean [SD], 241.59 [169.91] vs 330.97 [169.03] mg/dL, respectively; P = .02). Conclusions and Relevance: these results indicate that the screening of APOA5 and APOC3 genotypes may be useful to estimate changes in triglyceride concentrations during bexarotene treatment in patients with CTCL and also to identify the best candidates for bexarotene therapy based on the expected adverse effect profile

Faced with these situations that would demand a reinforcement in research and dissemination in diverse aspects of the humanities, from multiple perspectives, paradoxically an adverse panorama is drawn for the development and dissemination of humanistic knowledge, which concerns different factors. Some are related to the consideration of the area of knowledge itself, its organization within the scientific system, the questioning of its own limits, and the interaction with another knowledge. Considering current transition scenarios does not mean having to abandon old objectives, but it adds to the work conducted new objects of study closely related to current reality, such as: the informational revolution; the relations with the ecosystem and the environmental crisis; globalization; the intensification of human mobility and migration flows; the growing economic and social inequality; the frictions derived from the articulation of collective identities; the decolonization of discourses; demographic dynamics; integration of technological advances; and viability and support for alternative models of society.Peer reviewe

    Treatment with tocilizumab or corticosteroids for COVID-19 patients with hyperinflammatory state: a multicentre cohort study (SAM-COVID-19)

    Objectives: The objective of this study was to estimate the association between tocilizumab or corticosteroids and the risk of intubation or death in patients with coronavirus disease 19 (COVID-19) with a hyperinflammatory state according to clinical and laboratory parameters. Methods: A cohort study was performed in 60 Spanish hospitals including 778 patients with COVID-19 and clinical and laboratory data indicative of a hyperinflammatory state. Treatment was mainly with tocilizumab, an intermediate-high dose of corticosteroids (IHDC), a pulse dose of corticosteroids (PDC), combination therapy, or no treatment. Primary outcome was intubation or death; follow-up was 21 days. Propensity score-adjusted estimations using Cox regression (logistic regression if needed) were calculated. Propensity scores were used as confounders, matching variables and for the inverse probability of treatment weights (IPTWs). Results: In all, 88, 117, 78 and 151 patients treated with tocilizumab, IHDC, PDC, and combination therapy, respectively, were compared with 344 untreated patients. The primary endpoint occurred in 10 (11.4%), 27 (23.1%), 12 (15.4%), 40 (25.6%) and 69 (21.1%), respectively. The IPTW-based hazard ratios (odds ratio for combination therapy) for the primary endpoint were 0.32 (95%CI 0.22-0.47; p < 0.001) for tocilizumab, 0.82 (0.71-1.30; p 0.82) for IHDC, 0.61 (0.43-0.86; p 0.006) for PDC, and 1.17 (0.86-1.58; p 0.30) for combination therapy. Other applications of the propensity score provided similar results, but were not significant for PDC. Tocilizumab was also associated with lower hazard of death alone in IPTW analysis (0.07; 0.02-0.17; p < 0.001). Conclusions: Tocilizumab might be useful in COVID-19 patients with a hyperinflammatory state and should be prioritized for randomized trials in this situatio

El Congreso ofreció un espacio para la presentación, difusión e intercambio de importantes investigaciones nacionales e internacionales ante la comunidad universitaria que se dio cita en el encuentro. El uso de herramientas tecnológicas para la gestión de los trabajos de investigación como la plataforma Open Conference Systems y la web de presentación del Congreso http://citis.blog.ups.edu.ec/, hicieron de CITIS 2017 un verdadero referente entre los congresos que se desarrollaron en el país. La preocupación de nuestra Universidad, de presentar espacios que ayuden a generar nuevos y mejores cambios en la dimensión humana y social de nuestro entorno, hace que se persiga en cada edición del evento la presentación de trabajos con calidad creciente en cuanto a su producción científica.
